I thought about pairings I like, then decided to start with two pairings important to me.
Haruka Ten'ou/Michiru Kaiou (Pretty Soldier Sailor Moon): canon
This is an important pairing very me for being the first explicitly, unambiguously lesbian couple I had seen in fiction.
Parfait/Cacao (NG Knight Lamune & 40): strong subtext, Parfait/Lemon (NG Knight Lamune & 40 Fresh): one-sided canon
Not a good picture, but I like that all three of them are there. Anyway, all through the NG Knight Lamune & 40 series there was some subtext between Parfait (red-head) and Cacao (dark-skinned), but in the NG Knight Lamune & 40 Fresh O.V.A. Cacao, under the influence of strange powers, pasionately makes out with Cacao, the first explicit lesbian love-making scene I ever saw. Then a few episodes later, Parfait has a vision where Lemon (blonde) kisses her lovingly and admits to being in love with her, making it the first loli lesbian scene I ever saw. In the final episode, the real Lemon does porclaim that she is in love with Parfait as Parfait and Cacao depart.
I will think of other pairings later. What were first or important moments for you people? _________________

Hmm, first important pairings? Intersting topic. My important pairings would obviously also be yuri. I can't go as far back as you though; in fact, I probably didn't get really into the yuri until about five years ago.
Back in 2004, when I only watched anime legally, I fell in love with these two:
Eclair and Lumiere from Kiddy Grade - very strong subtext.
Sure, one of them had the body of a ten year old, but as the show progressed and obvious romantic undertones were presented between the two, culminating in one of those false symbolic kisses (resuscitation type thing), I found everything about it endearing.
I tried to find something simmilar to it after tha. I gave a slightly older series, Noir a purchase and was very pleased. These two were even more blatant toward the end:
Mirielle Boquet and Kirika Yumura of Noir - near cannon subtext.
Seeign the pure feelings of romantic attachment develop between these two hardened assassins in this beautiful series stuck with me a long time and firmly made me a yuri fan (I have a lesbian fetish, and had been enjoying lesbain porn and yuri hentai sincel ong before that though, so it was bound to happen anyway). From then on, I would skew any closeness between females in anime as being potential romantic energy.
A year or so later, my patience with the American anime industry's licensing schedules sort of fizzled out and I turned primarily to fansubbed anime. The very first show I downloaded fansubs of was Mai-HiME. These two stood out for me:
Natsuki Kuga and Shizuru Viola of Mai-HiME - cannon.
This is the first time I watched a series and the yuri was apparent and out front. Eclair/Lumiere and Mirielle/Kirika had all the elements of romance, but it never confirmaed anything. There was no doubt here that Shizuru wanted Natsuki in a romantic and even sexual sense. This was exciting for me.
I decided to actively search for yuri at that point. My search took me to my first ever actual yuri anime. As apparent as Kannazuki no Miko's faults are to me, these two held me captive in the end:
Chikane Himemiya and Himeko Kurusugawa of Kannazuki no Miko - cannon.
Here it was the two main characters, not some side characters like Shizuru and Natsuki, engaged in open yuri love. This was nice. It was enough to start my obsession. _________________

Fight! Iczer-One
Iczer 1/Nagisa Kanou: Very strong subtext
This was another pairing from early on. I remember seeing Fight! Iczer-One and thinking how the two heroines had to be lesbians; they seemed so close and on the verge of at least kissing, but it never really happens. Given that the only major male character in Iczer-One was the villain, it seemed Nagisa was definately into Iczer 1, and Iczer 1 had to like Nagisa in some way.
Colbalt/Sepia: Canon
Unlike the two above, Cobalt and Sepia were unambiguously a pair; the first time they are seen they are pasionately making love in bed, the first full-on lesbian sex scene I had ever seen. They are villains, but highly sympathetic ones who are only doing what they think is preserving their all-female race. _________________

Hmm, I've seen very few older anime, but Iczer-One sounds interesting enough.
I'm trying to think which random H title contained the first animated lesbian sex scene I saw, but it's escaping me entirely at the moment. _________________
